So then, what is the mechanic of Quirks? We are told in-universe that they are physical expressions of genetics. If that is the case, why would it be mind or soul defence, and not body?

Not necessarily. The same ‘adaptation’ argument from the early chapters would work. I.E. ‘Aizawa used his Quirk on Denki at the USJ, and his body adapted to block it.” They just never knew as this is the first time since then that it has been attempted.
