I can't believe no one has mentioned this yet - but there is an alternate ending you should watch, because they just green lit a sequel and it has considers that ending as canon. Most people agree it's the better version anyway; it only got changed because of some feedback from test screenings, and in the end they regretted the change they made. You would be very confused if you only ever saw this ending and saw a trailer for or watched the sequel at some point. Also, I don't know if you noticed the cracks looking like butterfly wings and antennae around the leader when everything went quiet and Will was "having a moment", but the whole butterfly thing makes more sense with the alternate ending too. My loose interpretation of the moment he has is that he realizes that just because something changes doesn't make it inherently bad or evil, and that all this time he's basically been trying to undo these creatures - from his perspective, help and cure them, but from theirs, he's kidnapping and killing them with essentially the goal of eradicating their species, effectively making him the "bad guy", which is one of the main reasons they changed the ending. They felt a heroic sacrifice to protect the cure would appeal to more people.

Y'all were also speculating about what "I am Legend" means exactly. I remember hearing at some point that it's something like, because Will is the odd one out among this city of "zombies", to them, he is like a legend that they'd speak of to each other, and watch out for, and live in fear of. Think about it from their point of view - your brethren just going missing and never come back, and you hear whispers that it may be because of this one being who is different from the rest of you and a threat to you all. They didn't even know where he lived; he was practically a living myth to them. And one more thing - when Will says all human behavior seems to have vanished, I think it was a combination of speculation and lost hope. It comes off less to me like he is stating absolute fact, and more like he has just convinced himself that there's nothing human left about them, just because HE doesn't see it in them - he just sees monsters. But the alternate ending gives more context for all of this, and shows that there was more to the behavior that he thought demonstrated the loss of basic survival instincts.
