I want to design a game where you play one out of three giantesses dedicated on taking over the world by conquering cities. Each giantess has her own motivation for doing so (This will be revealed later).

Cities are conquered by destroying buildings, vehicles and tormenting its civilians by crushing or devouring them. Once you terrorize the city enough, you will be given a choice to either enslave or decimate the city.

1) Decimation: enables you to grow to mega proportions and choose a body part to flatten the city. (Feet, hand, or butt). Decimated cities give you materials to customize your giantess.

2) Enslavement: By showing mercy, you submit all who live in the city, converting them into an army of worshippers. Worshipers devote their lives to their goddess by providing boosts and build new gear/outfits for the giantess using the material collected from other decimated cities.

Are those 2 options ready and, if so how much destruction is needed to activate them, and how do I destroy building?

Building destruction hasn't been implemented yet. I am waiting for Unreal engine to improve their destruction system to production ready. I have a small test demo using their current system but its too buggy to work on atm. Nevertheless, I will post a video of my test demo to help build a clearer image of what I intend to achieve with destruction.

Sounds cools, new outfits and customization is a pretty good carrot on a stick for game progression. Might want to add onto that a growth mechanic tying to game progression as well.

So far the growth mechanic is a gameplay feature that occurs as you destroy the city, the more you destroy the more you grow. Once a city suffers enough destruction, your will be promted with the choices mentioned above. Once you finish, you reset to your default giantess size. I thought about having the growth occur as the game progressed, but that would make the other game mechanics very hard to balance.
