I think it might be important to rewatch the story with the woman, and how the 3 cursed wombs were created (through the abortions she was forced to have). They're not curses or demons, but cursed wombs. Something in between human and curse basically. that's why Yuji feels upset about killing them, since they had physical bodies just like the children Mahito transfigured. So yes, the 3 are actual brothers from the same parents.

btw, part of why Gojo and Utahime knows there's a traitor is because of how Geto's team (Mahito, Hanami, etc) were able to find the students and knew gojo and others were going to be there in advance. Like how they planned for everyone else to be allowed inside the veil, except Gojo
