I think Johnny's right, the guy who was his friend, was the snitch, and that's why he killed him. The guy probably raised his son out of guilt. In my opinion, I think one of the baby's mothers (there were two women, only one gave birth obviously but I guess they were all going to escape together) was already becoming a hiruko, she must've had that disease we learned about in a prior episode... when they executed and hung the two women, one of them turned into the giant spider hiruko, which is why the baby has the same ice power as the spider. I like your other theories though. I haven't read the manga, so it's all theories here as well.

Another cool detail is that if you pay attention, the spider hiruko also had the red toy that juichi has, so its definitely one of the two women. Another theory I had is that they are the two girls that tokio saw kissing at the facility.
