NAMPT (Nicotinamide Phosphoribosyltransferase) is a key enzyme involved in the biosynthesis of NAD+ (Nicotinamide Adenine Dinucleotide), which is essential for various cellular processes, including energy metabolism, DNA repair, and regulation of gene expression. NAMPT plays a critical role in maintaining cellular health and function, and its activity is linked to numerous physiological and pathological processes.

As cells age, NAD+ levels tend to decline, which can lead to reduced mitochondrial function, increased oxidative stress, and accumulation of DNA damage. NAMPT's role in maintaining NAD+ levels makes it a key player in cellular aging processes. Enhancing NAMPT activity or boosting NAD+ levels through supplementation with precursors like NMN or nicotinamide riboside (NR) is being explored as a potential anti-aging strategy.

NAMPT also acts as a cytokine, known as Pre-B Cell Colony-Enhancing Factor (PBEF), which plays a role in inflammation and immune responses. It is involved in the regulation of immune cell function and has been implicated in various inflammatory diseases, including rheumatoid arthritis and sepsis.

NAMPT's role in maintaining NAD+ levels is also critical for brain health. NAD+ is involved in neuroprotection and cognitive function, and declines in NAD+ have been linked to neurodegenerative diseases like Alzheimer's and Parkinson's. Strategies to boost NAMPT activity and NAD+ levels are being studied as potential therapies for these conditions.
