To me there is a striking parallel between Dean with the mark of cain, and Sam with the demon blood back in season 4. How both boys act, the decisions they make, how they react to this dark force in/on them... I think this (and next season) is a massive empathy lesson for both boys- Dean for what Sam struggled with in having the demon blood, and Sam for how Dean felt watching Sam struggle with it.
