I’m not sure what to make of Damon saying he doesn’t want to be human again. If my only impression of him was based on the way he acts around the people who know him, I wouldn’t question him saying that he doesn’t want to be human. But in season 2, after Rose died, Damon had an emotional meltdown with a stranger in the road, where, before he killed her, he told her how much he missed being human, he missed it more than anything in the world. The very fact that he was speaking to a stranger who he then killed convinces me that, at the time, he sincerely did miss being human, because what would have been the point of lying to someone who didn’t know anything about him and who he killed immediately afterwards? So, in this episode, I can’t decide whether Damon actually has completely changed his mind that much in what is just about a single year in their timeline, or if he’s lying. I tend to lean towards lying, just because it seems unlikely that someone’s feelings could change that drastically in such a short period of time. And Damon does frequently go out of his way to keep people from seeing anything in him that could be perceived as a weakness or a vulnerability, and he’s expressed doubt as to whether the cure even exists, so if he does still miss being human I could totally believe he wouldn’t want people to know he wanted the cure until/unless he knows he can have it. Especially since he’s not convinced Elena’s feelings for him are real. I would imagine, in Damon’s mind, there could be nothing worse than for people to know he took the cure hoping for a human life with Elena only to have human Elena decide she didn’t want him anymore. I’m not 100% convinced he was lying, it’s more like 60%/40%, but if he was telling the truth and really doesn’t miss being human anymore, does think nothing could be more miserable, I have no theories about what could have made him change his mind so completely in just about a year.

I'm way late on this but I just started watching The Vampire Diaries on Netflix during Covid quarantine and stumbled onto your reaction channel on YouTube. I completely missed TVD when it was on TV. I'm really liking your reactions. Anyway, I think Damon said he didn't want to be human again because he still doesn't know if Elena truly loves him or not because of the whole sire bond business. He's protecting himself emotionally. After all, all he's heard for 150+ years from the women he loved is, "I love Stefan; it will always BE Stefan" and he can't bring himself to believe that the woman he's deeply in love with actually loves him.
