Although Margie Hines was the original voice of Betty, most people remember the voice of Mae Questel.  Questel did the bulk of the Betty Boop cartoon shorts from 1931 to 1938, also reprising the role 50 years later in Who Framed Roger Rabbit.  Child actress Shirley Temple is probably best known for her signature song, On the Good Ship Lollipop, featured in the 1934 film 'Bright Eyes'.  Questel, not known for singing talent at the time, recorded a cover of the song in Betty's voice...it sold over two million copies in the U.S. alone.

Again, not the first to voice the character, Questel did the bulk (over 25 years worth) of Olive Oyl from Popeye.  On the same token, Questel was supposed to voice Olive Oyl in Who Framed Roger Rabbit, but rights to the character (as well as Popeye and Bluto, among others) weren't obtained in time for filming.

In more modern times, Questel is remembered for her last non-voice actress role as Aunt Bethany in 1989's National Lampoon's Christmas Vacation, where she cites the Pledge of Allegiance at Christmas dinner.
