Hi there, 

Today I will show you the process to color a detailed colored sketch:

1. Final and fixed sketch checking the shadow to see how drawing works.

2. Clean the sketch adding details in clothes and char. I use a sharp brush with solid black.

3. Adding a solid base which will help to limit via clipping mask the adding of color, shadows and lights.

4. Shadow and light added to give volume to the char. Usually I make solid base and shadow/light in greyscales. It helps to have a better vision of the deepness of the char and the real degree of light and shadow you wanna get.

5. Solid color added from the char's palette.

6. Mixing layers using blending modes.

This is not the final version to render, after this step extra details are added, can be a background or something. About software same process can be done in Photoshop, SAI and other apps, personally I only work with Sketchbook pro.
