Each sprite for the game is traditionally drawn with sharpie marker and paper.  I'm heavy-handed, and tend to get my paws all over my pages, consequently this causes smudging for most of my ink/pencil drawings, so I find that sharpie marker works well for me.  [Side note, I thoroughly enjoy working with charcoal and clay, they are both fun and messy!]  When I work online, I use anatomy guides, art tutorials, and plenty of real life references.  I keep my trusty old light table nearby, an awesome force when combined with adobe photoshop in order to fix any weird lines, updating and make changes, or correct my mistakes to save a piece from being discarded or chucked into the shredder.   I play with layers in photoshop, my biggest problem is knowing when to stop working on a drawing.  My best asset is another person in the room, who has a different viewpoint.  I can ask them what they think of the drawing, and because their eyes are looking at the image for the first time instead of for 3+hours, I can trust their opinion, and can finally reason out a stopping point.  When finished, I make multiple save files, png as well as psd (never jpeg unless there's a reason for a low-res copy to share for business).
