A explanation about the scene with Zouken: Darnic Prestone Yggdmillenia (Vlad master) was a popular mage working in Transylvania at the time of the 3rd war he manged to convince the Nazi Party who were excavating magical treasures in hopes of winning the war. He then took the Nazi Magi who were helping him to Fuyuki and had them excavate the lesser grail which was in the form of a statue (As you saw at the beginning of the episode) as they didn't use homuncli until the 4th war. In this process they killed the Makiri (its implied that Zouken was killed) family as well as the Tohsaka and Einzberns who decided to retreat from the public eyes and practice magecraft without the grail.

So basically the events of fate/zero didn't happen, since the grail of this timeline is different from what you saw in f/zero and fate/SN.

So yeah like the others have been saying, in this timeline when Darnic stole the grail during the Third War Zouken died, which actually means its one of the best timelines for Sakura since there Matous died out. The sword that Shiro was using was originally a normal blade but Shakespeare/Caster buffed it to the level of a C rank Nobel Phantasm. And Yeah Shakespeare did put Berserker in a Genjutsu, he is basically a support Caster.
