The guide says Dalvie's shop is unlocked by 'doing the manor route', which I would take to be beating Edith however one save I have has beaten her but it is still locked. Does that require finishing Marin as well or something?

Nope, just beating Edith should be enough to unlock it, and all the other buildings in Transylvania. Check to make certain you've done so, otherwise it's possible your save is from an older version that's become defunct, in which case you may need to start a NG+ to fix it.

Edith is in the cell in the basement, but when I talk to Dalvie about unlocking the door she just says 'Why don't you return here after giving my wares a go?' which made me think of Marin.

As far as I know, all you have to do is talk to them; just pick the 'Talk' option and then ask about the locked door. Dalvie will ask about you. Either option works, and they'll be satisfied. (Note: Dalvie is identified with they/them pronouns, and their gender is yet-unknown. They may be non-binary, for all I know)

Confirmed that it unlocks after buying the living underwear from Marin, that should probably be mentioned in the guide.

What is it that activates the fairy grove? A new character I've made can go there, but the interaction spot in the middle is not there. The guide doesn't mention it, just talks about how to get there and what to do once the interaction is available.

Found that Cassie isn’t on the hand holding list. I know you are updating for r25. She was able to do hand holding in r24.1b if I remember correctly.
