To answer your questions: Kyuhyun and Min-ho joined the cast from season 3. In season 1 there was Hodong, Sugeun, Jiwon and Lee Seung-gi (actor). After Lee Seung-gi went to the military Jaehyun joined in season 2. I recommended season 4 first, because you can really see their dynamic from the start. Season 3 is also hilarious, but since they were still getting to know each other at the beginning, I thought this would be a good way for you to learn about them. As for the characters: they always have to dress up as one character from a Chinese tale called New Journey to the West. That's why the show is technically called that. But over the years they've added and changed the characters.

~Kyuhyun and Jaehyun are really good friends because of this show. ~Jaehyun is an actor but he wasn’t in the recent seasons of this show because he was in a “scandal” with his now ex wife. ~So to “replace him” they added Mino’s good friend P.O. from Block B
